Abstract

Both theoretical considerations and experimental data suggest potential neurotoxicity of intracerebral acting inhibitors of Catechol-O-Methyltransferase (COMT). Those drugs might induce increasing oxidative stress or higher levels of S-Adenosylmethionin, a COMT-cofactor with putative neurotoxicity. Subsequently, an increased formation of protoxins and toxins may result. Moreover, it has been shown that COMT-inhibitor tolcapone does impair mitochondrial energy production in vitro.
