The present study shows that invertase isoenzymes of Chenopodium rubrum are encoded by a small gene family comprising at least three members. In addition to the previously described cDNA ClN1, coding for an extracellular invertase, two Further invertase cDNA sequences, ClN2 and ClN3, have been cloned and characterised from Chenopodium rubrum. ClN2 encodes a putative acidic, vacuolar invertase, whereas the protein encoded by ClN3 has a unique combination of properties from both extracellular and vacuolar invertases cloned so far and thus represents a novel type of invertase isoenzyme. The effect of source-sink transition on the transcript levels of the different invertases has been investigated in suspension-culture cells. The mRNAs for the three isoenzymes are present in photoautotrophically grown suspension culture cells and are differentially regulated in response to induction of heterotrophic metabolism Whereas the mRNA concentration of ClN1 is induced by the addition of glucose, as shown before, the low transcript levels of both ClN2 and ClN3 are not affected. The physiological significance of the differential regulation pattern is discussed.
