Abstract

We investigated the changes in electric potential distribution of bilayer lipid membrane (BLM) due to phospolipase A2 activity and to the interaction of lipid hydrolysis products - lysolipids and fatty acids with BLM. To monitor the changes of boundary potential (DELTA(phib)) on BLM, we used method based on the compensation of inner membrane field revealed by the second harmonic of capacitive current. Xi-PotentiaI changes of liposomes were measured by the method of correlation spectroscopy; the potential distribution relative to the plane of hydrophobic ion adsorption was evaluated from the kinetic parameters of the hydrophobic ions (tetraphenylborate) transport across BLM. It was shown that during phospolipase A2 action each sumsequent phospholipid molecule is hydrolyzed such that the enzyme does not leave the membrane surface. The hydrolysis leads to the changes of the electric potential in different parts of the BLM surface layer. In the dioleoyl phosphatidylcholine membrane these changes are associated with the appearance of the lipid hydrolysis products - lysolipids and fatty acids.
